ABSTRACT

Abstract The aim of the present study is to assess the effects of selenium nanoparticles on the growth, hematology and nutrients digestibility of Labeorohita fingerlings. Fingerlings were fed with seven isocaloric sunflower meal-based diet supplemented with different concentrations of nanoparticles naming T1 to T7 (0, 0.5, 1, 1.5, 2, 2.5, and 3 mg/kg), with 5% wet body weight while chromic oxide was used as an indigestible marker. After experimentation for 90 days T3 treated group (1mg/kg -1Se-nano level) showed the best result in hematological parameters (WBC's 7.97 ×103mm-3, RBC's 2.98 ×106 mm-3 and Platelet count 67), nutrient digestibility (crude protein: 74%, ether extract: 76%, gross energy: 70%) and growth performance (weight gain 13.24 g, weight gain% 198, feed conversion ratio 1.5, survival rate 100%) as compared to the other treatment groups. Specific growth rates were found significantly higher in T5 than in other groups. The present study indicated positive effect of 1 mg/kg Se-nanoparticles on growth advancement, hematological parameters, and nutrients digestibility of L. rohita fingerlings.


Resumo O objetivo do presente estudo é avaliar os efeitos das nanopartículas de selênio no crescimento, hematologia e digestibilidade dos nutrientes de alevinos de Labeo rohita. Os alevinos foram alimentados com sete dietas isocalóricas à base de farinha de girassol suplementada com diferentes concentrações de nanopartículas, nomeando T1 a T7 (0, 0,5, 1, 1,5, 2, 2,5 e 3 mg / kg), com 5% do peso corporal úmido enquanto o óxido crômico foi usado como um marcador indigesto. Após a experimentação por 90 dias, o grupo tratado com T3 (nível 1mg / kg -1Se-nano) mostrou o melhor resultado em parâmetros hematológicos (WBC's 7,97 × 103mm-3, RBC's 2,98 × 106mm-3 e contagem de plaquetas 67), digestibilidade dos nutrientes (proteína bruta: 74%, extrato de éter: 76%, energia bruta: 70%) e desempenho de crescimento (ganho de peso 13,24 g, ganho de peso % 198, taxa de conversão alimentar 1,5, taxa de sobrevivência 100%) em comparação com os outros grupos de tratamento. As taxas de crescimento específicas foram encontradas significativamente mais altas em T5 do que em outros grupos. O presente estudo indicou efeito positivo de 1 mg / kg de nanopartículas de Se no avanço do crescimento, parâmetros hematológicos e digestibilidade de nutrientes de alevinos de L. rohita.

ABSTRACT

Background: Computer-aided repositioning of approved drugs is an increasingly popular strategy for the discovery of effective therapies. The potency of the newly repositioned drugs can be optimized using them as a component of an effective drug combination, thereby achieving the desired therapeutic effect at a lower and more tolerable drug concentration. Aim and Objectives: The aim of the study was to perform structure-based virtual screening and repurposing of FDA-approved drugs for the treatment of methicillin resistance by Staphylococcus aureus (SA) and perform an in vitro validation of the prediction. Materials and Methods: Following ethical clearance at the Department of Pharmacology and Therapeutics, College of Health Sciences, Usmanu Danfodiyo University Sokoto, molecular docking was performed against 5 validated protein targets involved in the development of methicillin resistance by SA and an in vitro validation of the prediction was done using 3 of the top-ranking drug candidates against methicillin-resistant vancomycin-susceptible strain of the pathogen (ATCC 43300). Results: Desmopressin and docetaxel, two of the 20 top-ranking repurposed drugs discovered through virtual screening, enhanced the inhibitory effect of oxacillin against the ATCC 43300 SA strain in a ratio-dependent manner, although each of the two drugs singly was only weakly effective against the bacterial strain. The standard drug, vancomycin (also among the top-scoring candidates), alone, was effective against ATCC 43300 strain and in combination with oxacillin, the two drugs produced a ratio-dependent synergistic effect against the bacterial strain. Conclusion: These findings suggest that oxacillin-based combinations with desmopressin, docetaxel, and the standard drug vancomycin, three of the 20 top-ranking drugs, at optimum ratios, may be beneficial in reversing the resistance of the ATCC 43300 SA strain to oxacillin, thus supporting the prediction of the molecular docking results.

ABSTRACT

Background: Intestinal helminthic infections are among the commonest infections worldwide. It often affects the poorest communities and has similar geographic distribution with malnutrition. Intestinal helminthic infection contributes to undernutrition through subtle reduction in digestion and absorption of food, chronic inflammation and loss of nutrients. The objective of this study is to determine the prevalence of intestinal helminthic infection and its relationship with nutritional status of primary school children in Gombe, Gombe State, Nigeria. Methodology: This was a cross sectional study of 350 pupils selected through multistage random sampling technique from 24 primary schools in Gombe, Gombe State, Nigeria from July 2018 to January 2019. Demographic information including age, gender, height, and weight were collected from each participant with a designed collection form. The data were analysed using SPSS version 24.0, and presented as frequency distribution and mean ± SD. The Chisquare test (with Odds ratio and 95% confidence interval) was used to test for association between prevalence of helminthiasis and factors such as gender, age group and school type. A p value of less than 0.05 was considered statistically significant at 95% confidence interval. Results: The prevalence of intestinal helminthic infection was 23.7% (83/350). Eighty (96.4%) of the 83 infected pupils were in public schools while only 3 (3.6%) were in private schools (p<0.001). The prevalence of helminthiasis was significantly higher in underweight pupils (34%, OR=2.113, p=0.0065)) and significantly lower (5.4%, OR=0.1637, p=0.0037) in overweight pupils while the prevalence was not significantly associated with normal weight (p=0.5482) or obesity (p=1.000). Conclusions: Intestinal helminthic infection is a public health problem in children with adverse significant relationship with nutritional status. Provision of toilet facilities in schools and periodic de-worming of pupils aimed at reducing loss of nutrients from intestinal helminthiasis are recommended.

ABSTRACT

Aims: This work is aimed at investigating physicochemical parameters and compositions of fatty acid, phospholipid and sterol of desert date (Balanites aegyptiaca) kernel and pulp. Study Design: Balanites aegyptiaca fruit is one of the oldest feed-stocks in Africa of which little or no attention has been given to it. The plant plays a diverse cultural and traditional role in different societies. Therefore, it is very important to explore more about the chemical composition of the kernel and pulp oils of Balanites aegyptiaca; since it is currently attracting considerable research interest as a result of its diverse beneficial properties. Methodology: The physicochemical parameters, fatty acids, phospholipids and phytosterols of B. aegyptiaca seed and pulp oils have been analyzed and compared with the standards and that of conventional oil for easy assessment of their suitability for nutritional and industrial applications. Results: The results of some physicochemical parameters of kernel and pulp oils were acid value (26.35 and 15.60 mg KOH/g), peroxide value (3.82 and 5.90 meq/kg), saponification value (162.40 and 198.60 mg KOH/g), iodine value (55.20 and 142.50 mg of I/100 g), specific gravity (0.93 and 0.92), kinematic viscosity (2.12 and 1.65 St) and refractive index (1.41 and 1.39), respectively. The most concentrated fatty acids were palmitic acid (14.53%) < linoleic acid (35.65%) < oleic acid (38.27%) for the kernel oil while that of pulp oil were linolenic acid (8.21%) < oleic acid (16.80%) < palmitic acid (32.70%) < linoleic acid (33.56%). Arachidic, behenic, lignoceric and myristic acids were all present in small quantities with none of them recording up to 1.0% in either of the samples. Caprylic, capric and lauric acids were determined but not detected in both oils. The fatty acid composition of kernel and pulp oils contained a healthy mixture of all the types of saturated and unsaturated fatty acids. The value of polyunsaturated/saturated index (P/S) which is associated to the impact on human health was higher in the pulp oil (2.47). Phosphatidylcholine had the highest content in both oils that is 75.99 and 25.88 mg/100 g, respectively. The total values of phytosterols for kernel and pulp oils were 85.00 and 9.87 mg/100 g, respectively. Conclusion: Balanites aegytiaca kernel and pulp oils have the potential to substitute several materials used in manufacturing oil in the chemical and pharmaceutical industries. However, in order to extend usage, these oils should be refined in order to improve the colour and taste.

ABSTRACT

Aims: Bovine tuberculosis (bTB) is a chronic infectious and contagious zoonotic disease of domestic, wild animals and humans. The disease occurs in a wide range of mammalian species and therefore, poses a public health threat. It also results in considerable economic losses in livestock production and carcass condemnation of infected cattle during meat inspection. This study was aimed at determining the prevalence of zoonotic bovine tuberculosis in slaughtered cattle, based on Post-Mortem (PM) meat inspection, Ziehl-Neelsen staining (ZN) and Polymerase Chain Reaction (PCR) techniques in abattoir and slaughter houses in Bauchi State, Nigeria. Place and Duration of Study: A cross-sectional abattoir based-study was conducted on 800 slaughtered cattle in the Northern, Central and Southern Zonal abattoirs of Bauchi State, Nigeria. This work was carried out between June-September, 2013. Study Design: Experimental. Methodology: One hundred and twenty (120) tissue samples from different organs were suspected to have bTB lesions at PM 15% (120/800). Out of the samples examined 35 (29.2%) were Acid-Fast Bacilli (AFB) positive; 10 (8.3%) of which were confirmed positive for M. bovis by the confirmatory Polymerase Chain Reaction (PCR). Results: The present study found the prevalence rates of 3.33% (4/120) and 5.00% (6/120) for males and females, respectively. This gave an overall prevalence of 8.33% for bTB (M. bovis) based on PCR. Bovine TB sex-specific rates were 10.00% (4/40) and 7.50% (6/80) by PCR, in males and females respectively. Female cattle also had a higher prevalence than male cattle but there was no statistically significant association (p>0.05, x2 = 0.218) between the presence of bTB in the tissues sampled and the sex of the cattle. There was a statistically significant association (p<0.05, x2 =7.002, OR=3.363) between detection of bTB in suspected tissues and the age of cattle. Using ZN, cattle aged six (6) years and above had the highest number of positive bTB cases 67.9% (31), while cattle aged 3-5 years old had the lowest 14.81% (4/27). PCR technique, revealed age-specific prevalence rate in cattle aged 6-8 and 9-11 years were 17.07% and 5.77%, respectively. Bauchi zonal abattoir had the highest number of suspected bTB cases (62.5%), followed by Katagum (26.7%) and Misau (10.8%). Conclusion: High infection rate of bTB was found among cattle sampled in the study area, with a significant prevalence in Bauchi metropolitan abattoir than the other two (2) slaughter houses (Katagum and Misau). This showed that the prevalence of bTB was higher in Bauchi metropolitan abattoir which supplies larger population of the state with beef. These findings also demonstrated that, there is urgent need for public health authorities in the state to intervene.

Objective: This study evaluated acute and sub-acute toxicities in rodents and microbial purity of a polyherbal formulation, Bobwell® popular among the natives for the management of diabetes mellitus (DM). It was prepared with unspecified quantities of the following plant materials viz. Gongronema latifolium. Garcinia kola, Vernonia amgydalina, Sphenocentrum jollyanum and Kigelia africana leaves. Materials and Methods: Microbial purity was evaluated on some bacterial and fungal organisms using appropriate diagnostic media. Toxicity of the polyherbal preparation was evaluated in Swiss albino mice by administering to the animals graded oral doses of the lyophilized preparation in the ranges of 1.0 to 20.0 g/kg body weight (bwt) and observed for changes. Wistar rats were also fed with different doses of the lyophilized formulation for 30 days and the effects on the biochemical profiles and haematological parameters were evaluated. Results: The purity evaluation test revealed presence of some bacterial organisms with the load within officially acceptable limits except Escherichia coli having a load of 1.50x102 cfu/ml while no fungal organisms were observed. The median acute toxicity value (LD50) of the polyherbal medicine was determined to be 15.2 g/kg bwt. There was significant increase (P ≤0.05) in the body weight of the animals treated with the highest dose of the formulation compared to the control. The biochemical parameters showed marked decrease in the plasma glucose level compared to the control. Increase in creatinine level was observed only in the animals that received the highest dose of the formulation while aspartate aminotransferase (AST) decreased significantly. On the other hand, alanine aminotransferase (ALT) exhibited significant increased (P ≤0.05) at the highest dose. The photomicrograph of hepatic tissue showed focal necro-inflammation around the portal hepatics. There was marked increase in the haemoglobin level and in the red blood cell (RBC) count at the highest doses. There was also significant increase in white blood cells (WBC). Conclusion: The high LD50 value indicated that the polyherbal preparations could be safe for use but its safety was negated by high presence of E coli load. Although the formulation showed good hypoglycaemic activity and beneficial effects on cardiovascular risk factors, at the highest dose, the formulation exhibited deleterious effect on the hepatic tissue.

ABSTRACT

The experiment was conducted to study the growth and development of pepper [Capsicum annum] grown on soil enriched with foliages from Leueaena Ieucocepha la, Ginelina arborea, Prosopis mimosa, Tectona grandis, Azadiracta indica and Damiella olverii. Top soil sample used for the experiment was collected from the University of Ilorin premises. Foliage from the selected tree species were collected, air dried and pulverized to a fine texture before mixing with soil. Dry pepper seeds were sown in the different soil mixture. The experiment was laid out as a Complete Randomized Design [CRD] with six treatments replicated thrice. Parameters evaluated include: Mean leaf area, Mean plant height and Mean leaf number. Samples of the air dried soil were taken before and after planting and used to determine total nitrogen, available phosphorus, organic matter content, potassium and soil pH. Results showed that the soil enriched with pruning from Azadiracta indica had the best performance in terms of the growth of pepper [Capsicum annum] as shown by the leaf area [2443.3 cm[2]], leaf number [55.0] and plant height [30.3cm] at 8 weeks after planting [SWAP], lire chemical composition of the soil also varied significantly among the treatments [P< 0.05] Prosopis mimosa had the best performance in terms of the pH amendment [6.6] and potassium content [2.8%] while Leucaena leucocephala had the best performance in term of nitrogen content [6.7%] and Organic matter content [5.4%]. Lastly, Daniella oliverii had the best performance in term of Phosphorus content [8.0]. In conclusion, pruning from Azcidiracta indica had the best performance in terms of the growth of Pepper

ABSTRACT

Dengue virus infection has become a global threat affecting around 100 countries in the world. Currently, there is no licensed antiviral agent available against dengue. Thus, there is a strong need to develop therapeutic strategies that can tackle this life threatening disease. RNA interference is an important and effective gene silencing process which degrades targeted RNA by a sequence specific process. Several studies have been conducted during the last decade to evaluate the efficiency of siRNA in inhibiting dengue virus replication. This review summarizes siRNAs as a therapeutic approach against dengue virus serotypes and concludes that siRNAs against virus and host genes can be next generation treatment of dengue virus infection.

ABSTRACT

A pot experiment was conducted to evaluate the efficacy of different oil cakes against the root knot nematode. Root knot nematodes are the major agricultural pest of wide range of crops worldwide. Chickpea, Cicer arietinum L., family Fabaceae, is a high yielding proteinaceous pulse crop with India being the leading producer. Root knot nematodes are one of the major biological constraint that reduces per capita yield of this pulse crops. In the present investigation, studies were made to determine the efficacy of different oil cakes viz. Azadirachta indica (Neem), Brassica campestris (Mustard) and Gossypium hirsutum (Cotton). Two doses (50 and 100 g) of each neem, mustard and cotton oil cakes were applied to reduce nematode infestation in chickpea. Results revealed that root gall development due to Meloidogyne incognita were significantly reduced in all the treatments and enhance all the plant growth characters of C.arietinum. Higher dose of neem oil cake was found to be most effective as compared to other treatments. Hence, it may be concluded that oil cakes are better substitute against nematicide for the effective control of root-knot nematode and reduce environmental hazards for ecologically safe environment.
